Learning outcomes

At the end of the module the student acquires methodological and application elements useful for integrating the knowledge obtained in the courses of Chemistry and Biochemistry.

Course contents

Kinetics and enzymatic regulation: dosage and spectrophotometric analysis of enzymes and their characteristics expressed by Km, Vmax, Kcat and Kcat / Km. Calculation of the Km and Vmax values by means of the double reciprocal diagram for lactate dehydrogenase.

Energy coupling and production of ATP for oxidative phosphorylation: measurement of respiration rate in isolated cells and mitochondria by oxigraphic analysis. Calculation of the rate of oxygen consumption in conditions of maximum speed of phosphorylation of ADP (state 3) and of "rest": state 4 of breathing. Evaluation of the degree of energy coupling by V (state3) / V (state4) ratio and phosphorylation efficiency by ATP / O calculatio

Teaching methods

The exercise is introduced by a short theoretical presentation on principles, methods and techniques subject to practical experience or by computer simulation

Assessment methods

The evaluation will take place in the context of the overall evaluation of the C.I. and in particular during the end of year interview

The exercises will be held in rooms equipped with computers
