18765 - Laboratory of Molecular Biology II

Academic Year 2009/2010

  • Teaching Mode: Traditional lectures
  • Campus: Bologna
  • Corso: First cycle degree programme (L) in Biotechnologies (cod. 0090)

Learning outcomes

Principles and practice of some of the basic techniques of molecular biology, such as heterologous expression and purification of recombinant proteins and protein/DNA interaction assays. Skills in analysis of experimental results and in their presentation.

Course contents

Expression of an Helicobacter pylori transcriptional regulator (Fur) in E.coli BL21(DE3) cells; purification of the protein through metal affinity cromatography; polyacrilamide gel electrophoresis (SDS-PAGE); dialysis and protein quantification with Bradford assay; electrophoretic mobility shift assay (EMSA); DNA electroblotting; Southern-blotting with non-radioactive DNA probes; ECL principles; basic photographic and imaging techniques.

Readings/Bibliography

Lab protocols and handouts.

Teaching methods

Each student will perform all the experiments individually; introductory lessons to principal themes; exercises will be made and discussed during the lab time.

Assessment methods

Written report; involvement and application during all the course will be considered.

Teaching tools

The course will be carried out in a fully equipped molecular biology lab; each student will be able to perform all the experiments individually.
