Learning outcomes
This course explores advanced methodologies for the structural and functional analysis of cells and human tissue using both imaging and spectroscopic techniques. Emphasis is placed on the integration of morphological and molecular data acquired through high-resolution microscopy and cutting-edge spectroscopy techniques, such as synchrotron-based X-ray methods. The course includes a practical computing activity focused on the post-processing and analysis of multidimensional datasets to extract meaningful biological insights.
Course contents
The course is organized into two integrated parts: 4 ECTS of lectures and 2 ECTS of computational laboratory activities.
The lectures are structured into four main blocks:
- From the biological question to measurable data
Experimental design, sample preparation, selection of controls, definition of quantitative endpoints, and main sources of variability or bias. - Physical principles of data generation
Interaction between electromagnetic radiation and matter, from the visible range to X-rays, with attention to contrast, resolution, signal, noise, dose and safety. - Imaging, spectroscopy and advanced platforms
Overview of the main methodological macro-areas, distinguishing conventional laboratory techniques from cutting-edge, limited-access approaches, such as synchrotron-based methods and cryo-EM. - From raw data to biological information
Principles of post-processing, quantitative analysis of multidimensional images and spectra, metadata management, uncertainty and reproducibility of workflows.
The laboratory component will allow students to apply, in a practical way, the concepts introduced during the lectures through computational exercises focused on the processing, analysis and interpretation of multidimensional datasets generated by imaging and spectroscopy techniques.

Written test aimed at assessing the student’s ability to apply the methods learned.
