Learning outcomes

Upon completion of the course, the student has a solid theoretical foundation for understanding the relationships between RNA structure and its regulatory and catalytic functions with possible therapeutic activities.

Course contents

RNA polymerase and classes of RNA. Structure of RNA polymerase II. Organization of class I, II and III promoters. General transcription factors and their functions. Structure of TBP in complex with DNA. Regulation of transcription. Upstream and inducible transcription factors. Enhancers. Maturation of transcripts. Secondary and tertiary structural motifs of DNA and RNA, examples of structured nucleic acids (riboswitches, ribozymes, long regulatory RNAs, ribonucleoprotein complexes in which catalytic activity is carried out by RNA subunits), recognition of metabolites and drugs by nucleic acids, use of small molecules for regulation of nucleic acid functions. The student will also learn the main experimental technologies used for the production, purification and structural characterization of RNA in vitro.
