Learning outcomes

At the end of the laboratory the student will be aware of the basis of Biochemistry with focus on enzyme kinetics and bioenergetics.

Course contents

Biochemical laboratory

- Kinetics and enzymatic regulation. Quantitative and spectrophotometric analysis of enzymes and their characteristics. Calculation of Km and Vmax.

- Energetic coupling and ATP production of oxidative phosphorylation. Measuring of respiration in cells and isolated mitochondria through oxygraphy. Calculation of oxygen consumption rate in state 3 and 4. Determination of the energetic coupling level with V(state 3)/V(state 4). Determination of the efficiency in oxidative phosphorylation through the ATP/O ratio.

Teaching methods

Instructor-led laboratory and bioinformatics activities.

Assessment methods

The laboratory work for Medicine and Surgery students (Channel A) contributes to achieving the educational objectives of the General Chemistry and Biochemistry and Metabolic Biochemistry modules. Student learning is assessed through a written exam in General Chemistry and Biochemistry and an oral exam in Metabolic Biochemistry; students can successfully pass these assessments by grasping the biochemical reasoning presented during the laboratory sessions.
