Learning outcomes

Basic knowledges about the main techniques necessary to perform a molecular analysis in the field of taxonomy, systematics and phylogeny in eukariots: isolation of nuclear, mitochondrial and plastidial DNA, electrophoresis, PCR amplification, bacterial cloning, restriction enzyme digestion; nuclear, mitochondrial and plastidial marker analyses, sequencing. Main software packages to be used for data analyses.

Course contents

analysis of moderately (f.i. ribosomal) or highly (f.i. satellite DNA) repeated sequences from animal genomes:PCR amplification of repeated sequences; cloning; positive clones recognition; sequencing. Data elaborations with Mega, DNAsp, MrBayes.
