Learning outcomes
The main objective is a basic knowledge on the techologies used to performed functional studies as a consequence of information obtained by genetic profile of cells. Specific knowledge on gene silecing through antisense oligonulecotides or siRNA and on gene trasnfection. Applications in medicine, advantages and limits for each tecnology will be discussed.
Course contents
1. Gene silecing by antisense oligonucletides, siRNA, ribozymes etc
2. Gene transfection and gene therapy
3. FISH, CGH and other technology to identify cheromosomal aberrations
4. Quantitative PCR.
An experimental part is also included. This will be related to:
1. Transient transfection of an Ewing sarcoma cell line with siRNA against IGF-IR;
2. Evaluation of the effectiveness of transfection
3. Isolation of RNA and reverse transcription in cDNA
4. Evaluation of the IGF-IR silencing by quantitative PCR.

Teaching methods
A practical part will be integrated by specific lessons on selected subjects. Experiments will be performed individually and the student can directly challenge himself with the application of what was theoretically exposed during the lessons.
Problems will be discussed and resolved meanwhile.
Assessment methods
A final written exam with several questions is forseen.
A detailed report on the experimental part is required for each student.
