SCIENTIFIC ACTIVITY - BRIEF DESCRIPTION
Lorenzo Bertin's research activity began in the late 1990s at the University of Bologna (Italy), focusing on the biodegradation of priority pollutants in actual-site PCB-contaminated soils by the development of bioprocesses in bioreactors with different nonconventional configurations. The culturing and employment of exogenous specialized bacteria was successfully proposed. Industrial wastewaters contaminated by non-ionic surfactants (polyethoxilated nonylphenols) and sewage sludge collected from secondary treatment plants of industrial wastewaters were then targeted for biodegradation studies, carried out under aerobic and anaerobic conditions, respectively.
The main topic of his Ph.D. project, carried out from 1999 to 2002, was the treatment and valorization of olive mill wastewaters (OMWs), i.e., the aqueous effluent resulting from olive oil production process, in aerobic and anaerobic fixed-bed biofilm reactors. In particular, he isolated two bacterial aerobic strains capable of biodegrading several polyphenolic compounds typically occurring in the wastewaters, which are responsible for the wastewater antimicrobial activity and phytotoxicity. The strains were successfully employed in the physiological condition of immobilized cells in the post-treatment of OMWs fed to anaerobic digestion processes. Anaerobic digestion processes were then investigated by developing an anaerobic biofilm column reactor, which was filled with granular activated carbon and which allowed significantly higher methane productivity with respect to conventional reactors.
OMWs and grape pomaces were then employed as feedstock for the development of multi-purpose biorefinery schemes dedicated to the recovery of polyphenols and the exploitation of the organic leftover for biopolymers (polyhydroxyalkanoates, PHAs) production. Selective, effective, and environmentally compatible solid phase extraction procedures, according to which both the adsorbent and the extraction solvent are recycled into the process, were defined for polyphenol or VFA separation and recovery. For the first time, the selective recovery of OMW polyphenols was coupled to the extensive conversion of the leftover organic matter into volatile fatty acids (VFAs), i.e., the most suitable substrate for aerobic mixed cultures capable of producing PHAs, by developing and optimizing an anaerobic acidogenic biofilm process.
From 2005, he designed, developed, and assessed unconventional anaerobic digestion (AD) processes carried out under methanogenic or dark fermentation conditions. Several organic matrices, among which mechanically sorted-organic fractions of municipal solid waste (MS-OFMSW), sewage sludge and agro-industrial effluents, have been tested as the process feedstock. In particular, he demonstrated that anaerobic microbial consortia capable of efficiently biomethanize polluted MS-OFMSW (recalcitrant to AD) can be obtained according to novel continuous or semicontinuous cultivation protocols, which allow the adaptation of consortia to contaminated organic fractions collected from unsorted MSW. Furthermore, an innovative two-stage AD process, which could combine the advantages of the simplicity of conventional processes with the high efficiency of multistep reactors, was designed and successfully developed. To this aim, the acidogenic stage was directly inserted into the methanogenic vessel according to a concentric design. The production of PHAs from agroindustrial residues has been pursued by the employment of pure bacterial cultures. To this aim, organic agro-industrial biowastes were anaerobically digested under acidogenic conditions, in order to obtain effluent rich in VFAs, which are suitable precursors for the biotechnological production of PHAs. Cupriavidus necator and Pseudomonas putida strains were tested as the main process biocatalysts in the frame of novel process schemes: strains were demonstrated to accumulate PHAs according to a dual-step approach, including a cell growth phase under balanced conditions and a consecutive PHA accumulation phase fed with VFAs. in particular, P. putida was forced to accumulate a specific amorfous and sticky PHA, i.e., polyhydroxyhexanoate (PHH), with peculiar alternative applications with repsect to the ones of conventional PHAs.
Finally, a novel research activity has been developing for the biological removal of sulphide from biogas by photofermentation processeses mediated by purple sulphur bacteria.
