R-loops and G-quadruplexes (G4) are non-B DNA structures that can be involved in transcription regulation and genome stability. G4s are four-stranded DNA structures composed of stacked guanine tetrads, and they are located in different critical positions of the human genome, notably at telomeres and in oncogene promoters. R-loop is a three-stranded nucleic acid structure, which consists of a DNA-RNA hybrid duplex and a displaced single stranded DNA. The aim of my project is the determination of R-loop profile and his variation in U2OS cells in the presence of G4-binders drugs, that can stabilize the G4 structure. The genome-wide profiling of R-loops is performed by DNA-RNA immunoprecipitation (DRIP) mediated by S9.6 antibody that recognizes RNA-DNA hybrid and whole genome sequencing.
